Experience in structural analysis
GlycoThera´s scientists have more than 25 years experience in the complete structural analysis of glycoprotein oligosaccharides using a broad spectrum of complementary procedures: including chromatographic sub-fractionation of the total glycans into individual chains and analysis by MS, MS-MS techniques (including derivatization methods) and NMR analysis for glycans and isolation of reference glycan structures for GMP release testing.
Analysis of specific oligosaccharide structures
Highest analytical standards using complementary analytical methods are used at GlycoThera´s laboratories to unequivocally corroborate oligosaccharide structural identities. Experience with > 80 recombinant glycoproteins analysed in our laboratories enables to identify very low amounts of e.g. potential antigenic structures or undesired motifs in recombinant glycoprotein therapeutics, e.g.:
-
Galα1-3Gal containing motifs
-
phosphorylated oligomannosidic glycans
-
isomeric oligomannosidic structures
-
hybrid-type glycans
-
peripheral (Lewis-type) fucose containing structures
-
O-acetylated sialic acids, N-glycolylneuraminic acid (Neu5Gc)
-
unusual truncated glycans
-
Galβ1-4 vs Galβ1-3 branching
-
unusual monosaccharide substitutions
Primary glycan structural analysis
N-glycan and O-glycan primary structure analysis comprises also:
-
glycosylation site occupancy (N- and O-glycosylation)
-
glycosylation site profile (LC-MS, MS-MS fragmentation)
-
glycoform mass (LC-ESI-MS)
-
monosaccharide composition
-
antennarity, isobaric structures and truncated oligosaccharide forms
-
rare oligosaccharide structures incl. potential antigenic glycan motifs
GlycoThera has set standards which have become mandatory for comparability studies and quality dossier submissions e.g. of biosimilar biopharmaceuticals.
